I need some clarification on how this works....Do the target machines have to
be manually configured in the BIOS to boot to PXE prior to the deploymet or
can this be done somehow with a deployment tool(s) like SIM,WAIK, or PE?

No deployment tool I know of can alter BIOS boot sequences to allow boot to
PXE. You would need to set this up on each target PC.
